You signed the petition to keep it around so you get to keep it around.

Forever.

Or until you go into Profile>Display>April Foolishness and set it to No Foolishness

So when next year's april fool's comes, will that setting disappear for a day or will the killjoys still be able to turn it off?

Foolishness will be "enforced." It won't be undoable with the box, but there'll probably be ways to get around it without it, like people did with some UserJS in 2011 and turning off javascript or using noscript or wrapping stuff in bbcode this year.
