There should be keys and gates and doors that act like colored versions but they have id. 1-99 I would be able to make a good tower defense level with these

There are 4 permanent gates/doors, one with values, as well as 7 timed gates/doors. You can also use portals and stuff. I don't see how it wouldn't be possible.
I like the idea of optionally adding ID's to keys and doors/gates for more control and more gate sets than just the current colors. Honestly, though, I can't picture what you mean by a tower defense game. If you mean a protect yourself game, I found you could make a "base" with multiple entry points, each guarded with a different colored gate and a matching key next to it on one side, the base side. For the attackers, you make a maze from the spawn to the base with each end connecting to a gate and the start guarded by a zombie only door so only attackers can go through. I know, right? I just started making one for my own purposes, but there are additions to the game that im waiting for to make it self functioning instead of owner monitoring. limited player gates, zombie infector block, etc. (you can look for the suggestions for more info)

I hope you realise that the largest part of a tower defense is building your own defense. Players cannot do this with just the things you suggested. They would need to be able to place their traps themselves, strategically. This would require some sort of edit area. I think what you are looking for is more of a capture the point game, but with traps, to compensate for the fact there is no form of PvP combat in EE.
